骨吸收标志物CTXI的检测
CTXI是I型胶原交联羧基端肽,Ⅰ型胶原由三条肽链组成,其中有两条α(Ⅰ)链,一条 α(Ⅱ)链,成熟的I型胶原分子的羧基端具有呈非螺旋的 3 条较短的肽链结构,CTXI是 I 型胶原在降解的时候从末端解离下来的肽段。由于胶原蛋白独特的三股超螺旋结构,性质十分稳定,一般的加工温度及短时间加热都不能使其分解,从而造成其消化吸收较困难,不易被人体充分利用。
I 型胶原主要分布于皮肤、骨、角膜、肌腱、肿瘤中,CTXI作为Ⅰ型胶原的降解指示剂,以完整的免疫原性蛋白形式进入血液中,不再进一步分解,故可作为骨吸收标志物,主要用于监测绝经后妇女和骨量减少个体的抗骨吸收治疗,评价患骨代谢疾病(如副甲状旁腺功能亢进、佩吉特病、骨营养不良),在局部骨溶解增加的状态下,如多发性骨髓瘤、溶骨性骨转移、类风湿性关节炎及固定术中,可观察到血清CTXI上升,同时CTXI对于乳腺癌骨转移早期诊断有一定价值。
由于这个端肽表达量较低(在人血清样本中浓度为142.6~1638.9pg/ml,在小鼠血清样本中浓度<545.8pg/ml ),故而研究难度较大,市面产品少。为了满足研究人员对其的研究需要,Cloud-Clone针对性的开发了CTXI端肽、抗体以及检测试剂盒等。端肽(SPA665Hu01、CPA665Hu21、CPA665Hu11、CPA665Hu31)、单克隆抗体(MAA665Hu21)和多克隆抗体(PAA665Hu01、LAA665Hu91和LAA665Hu71)的特异性都有实验数据支撑。检测试剂盒可定量测定人血清、血浆或其他相关生物液体中CTXI含量(CEA665Hu),Cloud-Clone还开发了小鼠(CEA665Mu)及大鼠(CEA665Ra)CTXI的检测试剂盒,以满足不同研究者的需要。
